Abstract

To know the effect of fetal bovine serum and change in calcium concentration on differentiation of cultured human kerationocytes, we cultured keratinocytes from 7 cases of burned patients under three conditions: 1.8mM Ca2+ concentration with or
without
10% fetal bovine serum(FBS), and 5mM Ca2+ concentration with 10% FBS.
Involucrin is a precursor of cornified envelope and a marker of differentiation of kerationocytes. We quantified the total intensity of fluorescence and the population of involucrin-positive cells by using the immunofluorescent staining of
involucrin
and flow cytometry, but there was no significant difference in three conditions(P>0.05).
It seems to be that FBS is not a remarkable stimulant in the differentiation of keratinocyte culture under Dulbecco's modified eagle medium, and increase of extracellular Ca2+ concentration to 5mM is not helpful in induction of differentiation.
To obtain more differentiated and thickened keratinocyte sheet, another methods should be investigated.
